Specificity (Detailed): Immunoblotting was performed using L-cells transfected with the tyrosinase gene and on four cell lines that express tyrosinase mRNA: SK-MEL-13, SK-MEL-19, SK-MEL-30 and SK-MEL-37 (1). Consistent with previous reports, a cluster of proteins ranging from 70-80 kDa was recognized by clone T311 in immunoblots of all five cell lines (1). An additional 55 kDa band was stained in blots from cell lines that are strongly reactive (SK-MEL-19) (1). Furthermore, immunoblots of untransfected L-cells and three cell lines that do not express tyrosinase mRNA (MZ2-MEL3.1, SK-MEL-187 and MZ2-MEL2.2) were negative with clone T311 (1). L-cells transfected with the tyrosinase-related protein 1, TRP-1(gp75), were also unreactive with clone T311 in immunofluorescence and immunoblotting assays, indicating this antibody does not cross-react with the melanosome-associated protein, TRP-1(gp75) (1).
